ABSTRACT

Los defectos congénitos constituyen causa de discapacidad física, psíquica y social, con un impacto en el incremento de los indicadores de morbilidad y mortalidad infantil. Pueden ser prevenibles en el nivel primario o preconcepcional. Con el objetivo de sistematizar los referentes teóricos relacionados con la embriogénesis de los defectos congénitos en función de su prevención, se realizó una búsqueda sobre el tema, consultándose 29 referencias bibliográficas. Se abordaron generalidades del desarrollo ontogénico, aspectos históricos de los defectos congénitos, su embriogénesis, etiología y definición. Asimismo, la prevención como estrategia de intervención en salud en diferentes niveles de actuación aplicados a los defectos congénitos. La etapa prenatal de la ontogenia -la más significativa- se divide a su vez en tres subetapas: la preembrionaria, la embrionaria y la fetal. La embrionaria, que transita entre la segunda y la octava semanas del desarrollo, es la más susceptible y vulnerable, y por tanto es el momento en el que, con mayor frecuencia, acontecen los errores en la embriogénesis, lo cual se traduce en el producto como un defecto congénito. La etiología de estos desórdenes de la ontogenia, obedece a factores genéticos, ambientales y multifactoriales. Existen tres niveles de prevención: preconcepcional, prenatal y posnatal; en el primero, se ponen en práctica diferentes acciones, y resulta vital la contribución de los médicos de la atención primaria de salud.


Birth defects cause physical, mental and social disabilities, with an impact on the increase of infantile morbidity and mortality indicators. They may be preventable at primary or preconception level. In order of systematize the theoretical references related to the embryogenesis of birth defects in function of their prevention, a search was carried out on the topic, consulting 29 bibliographic references. Generalities of ontogenic development, historical aspects of birth defects, their embryogenesis, etiology and definition are addressed. Also, prevention as strategy of health intervention at different levels of action applied to birth defects. The prenatal stage of ontogeny -the most significant one- is in turn divided into three sub-stages: pre-embryonic, embryonic, and fetal. The embryonic one, which passes between the second and eight weeks of development, is the most susceptible and vulnerable, and therefore it is the time when, most often, errors in embryogenesis occur, which translates into the product as a birth defect. The etiology of these ontogeny disorders is due to genetic, environmental and multifactorial factors. There are three prevention levels: pre-conceptional, pre-natal and post-natal; in the first, different actions are put into practice, and the contribution of the primary health care physicians is vital.

Introducción: Las poblaciones del coral Orbicella annularis han mostrado bajo reclutamiento en el Caribe. Uno de los cuellos de botella demográficos es la alta mortalidad en las primeras etapas de desarrollo. El conocimiento detallado del ciclo y las tasas de supervivencia de estas fases nos permitirá ayudar en la recuperación de la población y la restauración de los arrecifes. Objetivo: Describir la embriogénesis y estadios larvarios obtenidos por fertilización asistida y medir las tasas de asentamiento y supervivencia de las larvas en sustratos artificiales, antes de ser trasplantadas al arrecife. Métodos: Seis días después de la luna llena de septiembre de 2021, se recolectaron bolsas de gametos de ocho colonias de O. annularis en el Parque Nacional Natural Los Corales del Rosario y San Bernardo, Colombia, y se llevaron al laboratorio. Se realizó fecundación cruzada, se siguió el desarrollo embrionario y larvario hasta el asentamiento larval y se registró supervivencia hasta el día 41. Las larvas se mantuvieron en tres tanques con agua de mar filtrada con 126 sustratos marcados, previamente acondicionados con algas coralináceas costrosas. Luego, los sustratos se trasplantaron al arrecife. Resultados: El inicio del desarrollo embrionario ocurrió 1.11 hAF (horas después de la fertilización), cuando las células mostraron signos de la primera división, y duró hasta 104.59 hAF cuando comenzaron a metamorfosearse. El asentamiento de larvas se observó al sexto día AF. Veintiún días después de la fecundación se encontraron zooxantelas. La supervivencia de las larvas después del asentamiento fue de 27.5 %. Conclusión: En este primer esfuerzo de propagación sexual utilizando O. annularis en Colombia, 1.4 % de larvas competentes completaron todo el proceso de desarrollo. Aunque la tasa de supervivencia fue baja, estos resultados se suman a los esfuerzos de restauración de corales en el Caribe en los que se ayuda a las especies a aumentar la supervivencia de los corales en sus primeras etapas de desarrollo.


Introduction: Populations of the coral Orbicella annularis have shown low recruitment in the Caribbean. One of the demographic bottlenecks is the high mortality in the early stages of development. Detailed knowledge of the cycle and survival rates of these phases will allow us to assist in population recovery and reef restoration. Objective: To describe the embryogenesis and larval stages obtained by assisted fertilization and measure the settlement and survival rates of larvae on artificial substrates, before being outplanted to the reef. Methods: Six days after the full moon in September 2021, gamete bundles were collected from eight O. annularis colonies in Los Corales del Rosario and San Bernardo National Natural Park, Colombia and brought to the laboratory. Cross fertilization was carried out and embryonic and larval development were followed until larval settlement and survival was recorded until day 41. The larvae were kept in three tanks with filtered sea water with 126 tagged substrates, previously conditioned with crustose coralline algae. The substrates were then outplanted to the reef. Results: The onset of embryonic development occurred 1.11 hAF (hours after fertilization), when cells showed signs of the first cleavage, and lasted until 104.59 hAF when they began to metamorphose. Larvae settlement was observed on the sixth day AF. Twenty-one days after fertilization, zooxanthellae were found. Post-settlement larval survival was 27.5 %. Conclusions: In this first sexual propagation effort using O. annularis in Colombia, 1.4 % of competent larvae completed the entire development process. Although low survival rate, these results add to coral restoration efforts in the Caribbean in which species are assisted to increase the survival of corals in their early stages of development.

The hypothalamic-pituitary-thyroid axis gradually becomes mature at gestational age of 30~35 weeks.With the improvement of the treatment level of premature infants, the gestational age of surviving premature infants gradually decreases, and the thyroid axis of young premature infants is immature.Meanwhile, premature infants are more prone to systemic complications, such as ischemia and hypoxia, severe infection, etc., which aggravate the influence on thyroid.Clinically, more and more premature infants are found to be complicated with congenital hypothyroidism, temporary hypothyroxemia, hyperthyrotropin, delayed thyrotropin elevation, low T 3 syndrome and other problems.Abnormal thyroid function affects the outcome of the treatment of premature infant diseases.Early detection and early treatment is the key to improve the treatment, metabolism and the development of premature infant.At present, the timing of screening and treatment of premature thyroid disease is still controversial.In this review, the thyroid function and outcome of premature infants under different pathological conditions are summarized and analyzed to provide a reliable basis for rational selection of screening opportunities and treatment strategies for thyroid diseases in clinical practice.

ABSTRACT

Bisphenol A (BPA) is a monomer used in the production of polycarbonate, a polymer commonly found in plastics, epoxy resins and thermal papers. The presence of BPA in food, water, air and dust has been of great concern in recent years not only due to environmental and ecological issues but also because of its supposed risk to public health related to its mutagenic and carcinogenic potential. In this study we evaluated the toxicity of bisphenol A in zebrafish embryos (Danio rerio) and determined the 50% lethal concentration (LC50) of this chemical. BPA was used at concentrations ranging from 1 µM to 100 µM in E3 medium/0.5% dimethylsulfoxide (DMSO) from previously prepared stock solutions in 100% DMSO. Controls included embryos exposed only to E3 medium or supplemented with 0.5% DMSO. Camptothecin (CPT), a known inhibitor of cell proliferation was used as positive control at a concentration of 0.001 µM in E3 medium/0.5% DMSO. Adults zebrafish were placed for breeding a day before the experimental set up, then, viable embryos were collected and selected for use. Experiments were carried out in triplicates, according to specifications from Organization for Economic Cooperation and Development (OECD). One embryo/well (25 embryos per concentration) was distributed in 96 well microplates in presence or absence of the chemicals. The plates were kept in BOD incubators with a controlled temperature of 28.5 ºC and with photoperiod of 14 h light:10 h dark. After 24h, 48h, 72h and 96h exposure, the exposed embryos were evaluated according to the following parameters: mortality, coagulation, rate of heartbeat, hatching and presence of morphological abnormalities. Photography was obtained by photomicroscopy. Apoptosis was evaluated by DNA ladder assay. DNA was extracted by phenol:chloroform method and analyzed by 2% agarose gel electrophoresis. DNA fragments were visualized after ethidium bromide staining in ultraviolet transilluminator. The LC50 determined for BPA was 70 µM after 24 hours, 72 µM after 48 hours, 47 µM after 72 hours and 31 µM after 96 hours exposure. BPA induced morphological and physiological alterations such as yolk sac and pericardial edema, hatching delay or inhibition, spine deformation, decreasing in heartbeat rate and mortality. In conclusion, this study demonstrated that BPA induced marked malformations in zebrafish embryos at concentrations above 25 µM corroborating the current concerns related to the widespread presence of BPA in the air, food and water used by humans as well as in the bodily fluids and tissues.


Bisfenol A (BPA) é um monômero utilizado na produção de policarbonato, um polímero comumente encontrado em plásticos, resinas epóxi e papéis térmicos. A presença de BPA em alimentos, água, ar e poeira tem sido motivo de grande preocupação nos últimos anos, não só devido a questões ambientais e ecológicas, mas também ao suposto risco para a saúde pública relacionado ao seu potencial mutagênico e carcinogênico. Neste estudo avaliamos a toxicidade do bisfenol A em embriões de peixe-zebra (Danio rerio) e determinamos a concentração letal 50% (LC50) deste composto químico. O BPA foi usado na faixa de concentração entre 1 µM e 100µM em meio E3/0,5% de dimetilsulfóxido (DMSO), preparado a partir de soluções estoques em 100% DMSO. Os controles negativos incluíram embriões expostos apenas ao meio E3 ou suplementado com 0,5% DMSO. Camptotecina (CPT), um conhecido inibidor da proliferação celular, foi usado como controle positivo a uma concentração de 0,001 µM em meio E3/0,5% DMSO. Peixes-zebra adultos foram colocados para reprodução um dia antes da montagem experimental, em seguida, embriões viáveis foram coletados e selecionados para uso. Os experimentos foram realizados em triplicata, de acordo com as especificações da Organização para Cooperação e Desenvolvimento Econômico (OCDE). Um embrião/ poço (25 embriões por concentração) foi distribuído em microplacas de 96 poços na presença ou ausência dos compostos químicos. As placas foram mantidas em incubadoras BOD com temperatura controlada de 28,5 ºC e com fotoperíodo de 14h claro:10h escuro. Após 24h, 48h, 72h e 96h, os embriões expostos foram avaliados de acordo com os seguintes parâmetros: mortalidade, presença de coagulação, taxa do batimento cardíaco, eclosão e presença de anormalidades morfológicas. Fotografias foram obtidas por fotomicroscopia. A apoptose foi avaliada pelo ensaio de DNA ladder. O DNA foi extraído pelo método fenol:clorofórmio e analisado por eletroforese em gel de agarose a 2%. Fragmentos de DNA foram visualizadas após coloração com brometo de etídio em um transiluminador ultravioleta. A LC50 determinada para o BPA foi 70 µM após 24 horas, 72 µM após 48 horas, 47 µM após 72 horas e 31 µM após exposição por 96 horas. O BPA induziu alterações morfológicas e fisiológicas como edema de saco vitelino e edema pericárdico, atraso no tempo ou inibição da eclosão, deformação da coluna vertebral, diminuição da taxa de batimentos cardíacos e mortalidade. Em conclusão, este estudo demonstrou que o BPA induziu grande número de malformações em embriões de peixe-zebra em concentrações acima de 25 µM, corroborando as preocupações atuais relacionadas a presença generalizada do BPA no ar, alimento e água usados pelos seres humanos bem como nos fluidos e tecidos corporais.

ABSTRACT: Peach palm is a domesticated palm commercially important for the production of fruits and hearts of palm. Somatic embryogenesis, an effective technique for mass propagation, was successfully established for this species. Furthermore, a temporary immersion system improved plant regeneration. However, production can be further improved by understanding the peach palm's growth dynamic and modifications of culture media. The aims of this study were to evaluate the growth of plantlets cultured in different culture media in a temporary immersion system and to correlate the results with nutrient uptake during the growth period. Somatic embryo-derived young plantlets approximately 1 cm in length were cultivated for 12 weeks in a twin flask system containing MS, Y3 or N6 salts, Morel and Wetmore vitamins and 3% sucrose, with a monthly medium refreshment. Growth was measured and mineral analysis of the plantlets was carried out after 12 weeks of culture. The Y3 and MS salts were the most appropriate for the plant growth. Number of roots was 52.52% higher and the root size was 40.42% between the N6 and MS medium and the root number in Y3 medium was 37.74% greater than in MS medium, which is important for post acclimatization survival. K and Na are important elements for peach palm. N is not required at such a high concentration as in Murashige and Skoog formulation. The Chu (N6) medium did not generate high quality plantlets, possibly due to the absence of some micronutrients, like Mo, Cu and Co.


RESUMO: A pupunheira é uma palmeira comercialmente importante para a produção de palmito. A embriogênese somática, técnica efetiva para propagação massal, foi estabelecida com sucesso para essa espécie. Além disso, um sistema de imersão temporária aumentou a regeneração de plantas. Entretanto, a produção pode ser melhorada através da compreensão da dinâmica de crescimento e modificações do meio de cultura. O estudo objetivou avaliar o crescimento de plantas em diferentes meios de cultura em um sistema de imersão temporária e correlacionar os resultados com a absorção de nutrientes durante o período de crescimento. Plantas derivadas de embriões somáticos, com aproximadamente 1 cm de comprimento, foram cultivadas por 12 semanas em um sistema tipo frasco gêmeo contendo sais do MS, Y3 ou N6, vitaminas de Morel e Wetmore e 3% de sacarose, com renovação mensal do meio de cultura. O crescimento e os teores de nutrientes nas plantas foram determinados após 12 semanas de cultivo. Os sais do MS e Y3 foram os mais apropriados para o crescimento vegetal. O número e comprimento de raízes foi 52,52% e 40,42% maior no meio MS do que no meio N6, respectivamente, e o número de raízes no meio Y3 foi 37,74% maior que no meio MS, o que é importante para a sobrevivência após a aclimatização. K e Na foram os nutrientes mais importantes para a pupunheira. O N não foi requerido em altas concentrações como verificado na formulação do meio Murashige e Skoog. O meio de Chu (N6) não gerou plantas de boa qualidade, possivelmente devido à ausência dos micronutrientes Mo, Cu e Co.

Introduction: Rehabilitation of hermatypic coral species that have declined in the Caribbean in recent decades is a priority. Production of sexual recruits is considered the best restoration method to aid affected populations. Objective: To gain knowledge of early life stages of Orbicella faveolata and to enhance production of new sexual recruits. Methods: Gamete bundles from the coral species O. faveolata were collected over two years (2018 and 2019) from Los Corales del Rosario y de San Bernardo Natural National Park, Cartagena, Colombia. Assisted fertilization, larval rearing, settlement (onto crustose coralline algae, CCA) and post settlement survival in laboratory conditions were monitored. Results: Embryonic and larval development were documented over 55 hours after the first cleavage, when larvae were fully developed and started pre-settlement behavior. Settlement began 7 days after first cleavage and after 37 days polyps had acquired zooxanthellae. Larval settlement was higher on Lythophyllum congestum and Titanoderma prototypum than in response to Porolithon pachydermum, Neogoniolithon sp., Hydrolithon sp., and Lythophyllum sp. Larvae did not settle on dead coral or on the negative control (sterilized seawater). After the first week post settlement survival was 59 % amongst O. faveolata recruits. During the second week, survival dropped to 42 %, and was further reduced to 0 % at the end of the third week. Conclusions: O. faveolata larvae require cues from certain CCA species to settle, they do not settle in absence of CCA. Increased larvae availability is possible through assisted fertilization in the laboratory, however, due to the high mortality in early post-settlement phases, additional research needs to be conducted in order to scale up larvae production and improve understanding of the cues that enhance settlement and the factors which cause post-settlement mortality.


Introducción: La rehabilitación de las especies de corales hermatípicos del Caribe que han disminuido en las últimas décadas es una prioridad. La producción de reclutas sexuales se considera el mejor método de restauración para ayudar a las poblaciones afectadas. Objetivo: Obtener conocimiento de las primeras etapas de la vida de O. faveolata y mejorar la producción de nuevos reclutas sexuales. Métodos: Por dos años (2018 y 2019), seis días después de luna llena en septiembre, se recolectaron paquetes gaméticos en arrecifes del Parque Nacional Natural Los Corales del Rosario y de San Bernardo, Cartagena, Colombia. Se siguió la fertilización asistida, la cría de larvas, el asentamiento y la supervivencia posterior al asentamiento en algas coralinas costrosas (ACC) en condiciones de laboratorio. Resultados: El desarrollo de embriones y larvas se documenta a lo largo de 55 h después del primer clivaje, cuando la larva está desarrollada completamente y comenzó el comportamiento previo al asentamiento. El asentamiento comienza 7 días después del primer clivaje y 37 días después, la mayoría de los pólipos presentan zooxantelas. El asentamiento larval fue más alto en Lythophyllum congestum y Titanoderma prototypum que en respuesta a Porolithon pachydermum, Neogoniolithon sp., Hydrolithon sp., y Lythophyllum sp. No hubo asentamiento sobre coral muerto ni en el control negativo (agua de mar esterilizada). La supervivencia bajó de un 59 % en la primera semana después del asentamiento, a 42 % durante la segunda semana y 0 % para el final de la tercera semana. Conclusiones: Las larvas de O. faveolata requieren señales de ciertas especies de ACC para asentarse, ellas no se asientan en ausencia de ACC. La disponibilidad de larvas es posible mediante la fertilización asistida en laboratorio. Sin embargo, debido a la alta mortalidad en las primeras fases posteriores al asentamiento, queda mucho por hacer para aumentar la producción de larvas y mejorar nuestro conocimiento y comprensión de las señales que mejoran el asentamiento y las que previenen o inhiben la supervivencia del recluta.

RESUMO Objetivo Elucidar se a suplementação com ácido fólico pouco antes da concepção e/ou durante a gestação pode estar realmente atrelado ao desenvolvimento do transtorno do espectro autista (TEA). Metódos Foi realizada uma revisão de literatura em base de dados, nos idiomas português e inglês, durante o período de novembro de 2017 até abril de 2018, com ênfase nas publicações mais recentes. Resultados Do total de 174 artigos, 87 compuseram este trabalho. Pesquisas apontam que o aumento dos casos de TEA se deve ao fato de que mais fatores genéticos estejam implicados na etiopatogênese neural. No entanto, a grande maioria dos artigos ressalta com maior precisão que há mais efeitos benéficos do uso de ácido fólico antes da concepção e durante a gestação na prevenção do TEA, assim como de outras anormalidades relacionadas aos defeitos do tubo neural. Conclusão Quando se analisa o risco-benefício da suplementação com ácido fólico nas doses recomendadas, 0,4 a 0,8 mg/dia, conclui-se que os benefícios sobrepujam os possíveis riscos de desenvolver o TEA.


ABSTRACT Objective Elucidating whether supplementation with folic acid shortly before conception and/or during pregnancy may actually be linked to the development of Autistic Spectrum Disorder (ASD). Methods A literature review was conducted in the Portuguese and English languages during the period from November 2017 to April 2018, with emphasis on the most recent publications. Results Of the total of 174 articles, 87 compose this work. Research indicates that the increase in ASD cases should take into account the fact that more genetic factors are implicated in neural pathogenesis. However, a large majority of articles point out that there are more beneficial effects of using folic acid before application and during pregnancy in the prevention of ASD, as well as other abnormalities related to neural tube defects. Conclusion When analyzing the risk-benefit of folic acid supplementation at the recommended doses, 0.4 to 0.8 mg/day, it is concluded that the benefits outweigh the possible risks of developing ASD.

Background: Plant gene homologs that control cell differentiation can be used as biotechnological tools to study the in vitro cell proliferation competence of tissue culture-recalcitrant species such as peppers. It has been demonstrated that SERK1 homologs enhance embryogenic competence when overexpressed in transformed tissues; therefore, cloning of a pepper SERK1 homolog was performed to further evaluate its biotechnological potential. Results: A Capsicum chinense SERK full-length cDNA (CchSERK1) was cloned and characterized at the molecular level. Its deduced amino acid sequence exhibits high identity with sequences annotated as SERK1 and predicted-SERK2 homologs in the genomes of the Capsicum annuum CM-334 and Zunla-1 varieties, respectively, and with SERK1 homologs from members of the Solanaceae family. Transcription of CchSERK1 in plant tissues, measured by quantitative RT-PCR, was higher in stems, flowers, and roots but lower in leaves and floral primordia. During seed development, CchSERK1 was transcribed in all zygotic stages, with higher expression at 14 days post anthesis. During somatic embryogenesis, CchSERK1 was transcribed at all differentiation stages, with a high increment in the heart stage and lower levels at the torpedo/cotyledonal stages. Conclusion: DNA sequence alignments and gene expression patterns suggest that CchSERK1 is the C. chinense SERK1 homolog. Significant levels of CchSERK1 transcripts were found in tissues with cell differentiation activities such as vascular axes and during the development of zygotic and somatic embryos. These results suggest that CchSERK1 might have regulatory functions in cell differentiation and could be used as a biotechnological tool to study the recalcitrance of peppers to proliferate in vitro.

ABSTRACT

Somatic embryos (SE) of habanero pepper (Capsicum chinense Jacq.) represent persistent deformations in the shoot apical meristem (SAM), which inhibits their capacity to form organs and subsequently plants. In dicotyledonous plants, SAM is formed in the apex, between cotyledons and it plays a central role in postembryonic shoot organ formation. Based on the previous knowledge on the role of some families of gene in the formation, organization and maintenance of the SAM, the expression patterns of WUS, WOX2, NAM, STM, PIN1 and PIN7 genes were analysed, which would allow us to elucidate the possible implication of these genes in SAM deformations in the SE of C. chinense. The results show that the expression patterns of STM and PIN1 in the SE were completely opposite to the respective expression pattern obtained in zygotic embryos (ZE). Moreover, NAM and PIN7 showed an over accumulation of transcripts in SE, compared with ZE. This is the first time in the genus Capsicum that alterations in the expression pattern of key genes of the SE development are reported, as well as its possible implication in the persistent deformations of the SAM.

Over the past few decades, many reports were published in scientific journals describing medicinal properties of Ficusglomerata (FG). However, its effects on embryonic development and its safety characteristics have not been studied.The purpose of this investigation was to determine lethal concentration 50 (LC50) and study the effect of aqueousextract of FG leaf (AEFG) on developmental abnormalities in zebrafish embryos. LC50 value of AEFG was calculatedby using probit analysis. Effect on percentage hatchability, heartbeat rate, total body length, and developmentalmorphological abnormalities, i.e., delayed growth, abnormal movement, tail detachment, abnormal head-trunkangle, scoliosis/flexure, and yolk sac edema were recorded. AEFG revealed LC50 of 239.88 ppm. The result showeda significant reduction in percent hatchability (p < 0.05), heartbeat rate (p < 0.001), total body length (p < 0.001),and developmental morphological abnormalities in the embryos treated with AEFG. This research can be used inconsidering the safety of an AEFG extract for their use during pre-conception or early pregnancy period.

ABSTRACT

Aim: To study the effect of various plant growth regulators (PGRs) for induction of somatic embryogenesis and plantlet formation from cotyledon and leaflet explants in S. nigrum (night shade) an important medicinal plant used in treatment of digestive problems and skin infections. Place and Duration of Study: Department of Biotechnology, Kakatiya university, Warangal. Telangana, India, 3 years. Methodology: Cotyledon (0.8 cm2) and leaflet explants (0.8-1.0 cm2) from 3 week and 4 week old were cultured on MS medium supplemented with 30 g/L sucrose along with different concentrations of 0.5 mg/L BAP+NAA (0.5 – 6.0 mg/L) . Results: Maximum percentage of somatic embryogenesis was observed in cotyledon(89%) and leaf (98%) explants on MS medium augmented with 0.5mg/L BAP in combination with 2.0 mg/L NAA whereas the highest number of somatic embryos per explant (86 ± 0.19) was formed in leaflet explant. Conclusion: Somatic embryogenesis was induced from both cotyledon and leaf explants. Since it is threatened and medicinally important species S. nigrum, the present protocol can be used for its conservation and genetic transformation experiments.

ABSTRACT

Sonic Hedgehog gene (SHH) plays a vital role in embryogenesis through its secreted protein sonic hedgehog protein (Shh). During embryogenesis, Shh acts as a morphogen controlling proximal and distant signaling in the specific development of tissue lineages, patterning, regulation of cell proliferation and suppression of tissue apoptosis. Shh also exerts its role in odontogenesis by determining the site of tooth bud formation, in tooth morphogenesis and root formation. The difference in the specific development of a region by Shh can be explained by its [a] 'Spatial gradient [b] the 'form' [c] Concentration gradient and [d] Temporal gradient. Shh signaling pathway has an extracellular and an intracellular component. A disruption of Shh pathway contributes to tumorigenesis of several cell types including those arising from odontogenic structures. This article reviews Shh from its formation in embryonic stages, its role in development and odontogenesis, to its reactivation in tumorigenesis and in specific to odontogenic pathologies.

ABSTRACT

Campomanesia adamantiumis a native plant species of Brazilian Cerrado with diverse economic potential and great medicinal importance. Its sexual propagation is impaired by the recalcitrance of its seeds, which prevents effective and profitable propagation. With the purpose of establishing commercial crops and minimizing the extractive use of vegetal resources, the aim of the present study was to induce embryogenic calli in nodal segments of gabirobeira, and to determine and characterize their embryogenic phase through the establishment of a growth curve based on cellular characteristics. Calli were induced using nodal segments inoculated in WPM culture medium without the addition of hormones (control) and with different concentrations of 2,4-D, IAA, IBA, NAA or picloram. Cytochemical and SEM analyses revealed cellular characteristics of the formation of meristematic centers that indicated 4.14 μM of picloram to be the best treatment for induction of embryogenic calli, and demonstrating their embryogenic potential. The treatment was used to establish a callus growth curve, from which it was inferred that calli should be transferred to new culture media on the 28thday to maintain cell viability.

ABSTRACT

BACKGROUND: To assess the expression of T-box transcription factor 4 (TBX4) during the anorectal development in normal and ethylenethiourea (ETU)-induced anorectal malformations (ARM) rat embryos. METHODS: Anorectal malformations was induced by ETU on the 10th gestational day (E10) in rat embryos. Spatiotemporal expression of TBX4 was evaluated in normal (n = 490) and ETU-induced ARM rat embryos (n = 455) from E13 to E16 by immunohistochemical staining, Western blot analysis and real-time RT-PCR. RESULTS: In the normal embryos, immunohistochemical staining revealed that TBX4 expression was detected in the epithelium of hindgut and urorectal septum (URS) on E13. TBX4-immunopositive cells were increased significantly in the epithelium of hindgut and URS, the future anal orifice part of cloacal membrane on E14. On E15, abundant stained cells were observed in the rectum, URS and dorsal cloacal membrane and the expression of positive cells reached its peak. On E16, only sporadic positive cells were distributed in the epithelium of the distal rectum. In the ARM embryos, the hindgut/rectum, URS and dorsal cloacal membrane were faint for TBX4 immunohistochemical staining. In the normal group, TBX4 protein and mRNA expression showed time-dependent changes in the hindgut/rectum from E13 to E16 on Western blot and real-time RT-PCR. On E13 and E15, the expression level of TBX4 mRNA in the ARM group was significantly lower than that in the normal group (P < 0.05). On E15, the expression level of TBX4 protein in the ARM group was significantly lower than that in the normal group (P < 0.05). CONCLUSIONS: The expression of TBX4 was downregulated in ETU-induced ARM embryos, which may play important roles in the pathogenesis of anorectal development.

ABSTRACT: The aim of the present study was to induce the formation of somatic embryos in protocorms from Phalaenopsis Classic Spotted Pink hybrids at two physiological maturation stages, namely: 80 and 120 days after seed inoculation (DASI). Protocorms were inoculated in ½ MS medium supplemented with 0.1 mg L-1 ANA and 3 mg L-1 TDZ. Protocorms were inoculated 120 days after sowing were more developed at the 15th cultivation day due to the formation of pro-embryogenic structures. It was possible seeing the formation of globular- and torpedo-stage somatic embryos at the 30th day of cultivation in somatic embryogenesis (SE) induction medium. The protocorms inoculated at the 80th DASI did not formed somatic embryos; they oxidized 20 days after cultivation in SE-induction medium. The formation of somatic embryos happened directly on the explant, thus characterizing a direct somatic embryogenesis. The embryos converted into plants when the somatic embryos were transferred to the nutrient medium containing no growth regulator. Therefore, it was concluded that the somatic embryos induction from protocorms 120 days after sowing was positive, since the embryos were able to become plants and presented vegetative organs with morphological traits similar to those of the matrix plant.


RESUMO: O presente estudo buscou induzir a formação de embriões somáticos em protocormos do híbrido Phalaenopsis Classic Spotted Pink em dois estádios de maturação fisiológica aos 80 e 120 dias após a inoculação das sementes (DAIS). Os protocormos foram inoculados em meio ½ MS suplementado com 0,1 mg L-1 de ANA e 3 mg L-1 de TDZ. Aos 15 dias de cultivo os protocormos, inoculados aos 120 dias após a semeadura, apresentavam estádio mais desenvolvido, apresentando formação de estruturas pró-embriogênicas. Aos 30 dias de cultivo em meio de indução de ES observou-se a formação de embriões somáticos na fase globular e torpedo. Os protocormos inoculados com 80 DAIS não evoluíram para a formação de embriões somáticos, ocorrendo a oxidação destes aos 20 dias após o cultivo em meio de indução de ES. A formação dos embriões somáticos ocorreu diretamente no explante, caracterizando uma embriogênese somática direta. Quando os embriões somáticos foram transferidos para o meio nutritivo sem regulador de crescimento, houve a conversão em plantas. Diante disso, conclui-se que a indução de embriões somáticos a partir de protocormos com 120 dias após a semeadura, foi positiva, em que os embriões obtidos apresentaram competência em converter-se em plantas, apresentando os órgãos vegetativos com características morfológicas satisfatórias.

RESUMEN Eremophilus mutisii, pez catalogado en estado como vulnerable, que por su importancia sociocultural y ecológica genera retos investigativos, uno de ellos, conocer sus características histológicas del desarrollo embrionario, desde la fecundación hasta la eclosión. Para ello, se obtuvieron embriones de individuos adultos y maduros sexualmente, capturados en la represa del Sisga, ubicada en el municipio de Chocontá, Cundinamarca, inducidos sexualmente con extracto pituitario de carpa. Los ovocitos y el semen fueron obtenidos por presión abdominal 24 horas después de la última dosis, a 14±1°C. La toma de muestras, se realizó cada 60 minutos, durante las primeras doce horas post-fertilización (HPF); a partir de aquí y hasta la eclosión, las muestras fueron fijadas en formol Bufferado al 4%, cada 6 horas y sometidas a rutina de histología. Los resultados mostraron que los óvulos fertilizados presentaron forma esférica, color verde claro y diámetro promedio de 1270µm. El primer clivaje, se observó a las 2HPF; la blastulación inició a las 9HPF; los movimientos epibólicos y la gastrulación, se observaron desde la 12HPF hasta la 30HPF, con el cierre del blastoporo. A las 60HPF, se evidenció el desprendimiento de la cola del saco vitelino y a las 66HPF, se dio inicio a los primeros movimientos autónomos. Finalmente, a las 72HPF inició la eclosión, mostrando las larvas ausencia de pigmentación y evidenciándose un tracto digestivo rudimentario, sin abertura bucal y anal. Se observó la presencia de abundante saco vitelino, con movimientos verticales continuos de la larva.


ABSTRACT Eremophilus mutisii, due to its vulnerable condition and its potential for fish farming, generates research challenges; one of them is to know the embryonic changes of the E. mutisii based on morphological and histological features from fertilization to hatching. The embryos were obtained from adult and sexually mature individuals of the Capitán de la Sabana (Eremophilus mutisii), sexually induced with carp pituitary extract. Oocytes and semen were obtained by abdominal pressure 24 hours after the last dose at 14±1°C. Sampling was performed every 60 minutes for the first twelve hours´ post-fertilization (HPF); from here on at hatching, the samples were fixed every 6 hours. The embryos were fixed in 4% formalin for observing the morphological features and for the histological studies. Results revealed that the fertilized eggs had spherical shape, light green color and average diameter of 1270µm. The first cleavage at 2 HPF was observed, the blastulation began at 9 HPF. Epibolic movements and gastrulation were observed from 12 HPF to 30 HPF with blastopore closure. At 60 HPF the yolk sac tail detachment was evident and at 66 HPF the first autonomous movements began. Finally, at 72 HPF, the hatching began, showing absence of pigmentation and evidencing a rudimentary digestive tract without oral and anal opening. It was observed the presence of abundant yolk sac with continuous vertical movements of the larva.

Background: Somatic embryogenesis receptor-like kinase 1 (SERK1) is a cell membrane receptor active in different plant tissues and involved in cell differentiation activities including somatic embryogenesis. The identification of promoter elements responsible for SERK1 expression during the onset of somatic embryogenesis can be useful to understand the molecular regulation of the cell-to embryo transition, and these promoter elements represent biotechnological tools in plant organ tissue culture. Results: A −1,620 bp DNA sequence located upstream of the Coffea canephora SERK1 gene homologue (CcSERK1) was isolated, and then, different segments containing key response elements (REs) for somatic embryogenesis onset and development were fused to the uidA (encoding a ß-glucuronidase, GUS) reporter gene to evaluate its expression in transgenic leaf explants. DNA segments of −1,620 and −1048 bp in length directed uidA expression with patterns in leaf explants similar to those occurring during somatic embryogenesis. When a −792-bp fragment was used, uidA expression disappeared only in leaf explants and pro-embryogenic mass but persisted in developing embryos. No uidA expression was detected in any embryogenic stage when a −618-bp fragment was used. Conclusion: DNA deletions showed that a −1048-bp sequence located upstream of the CcSERK1 gene is sufficient to direct gene expression during the onset and the development of C. canephora somatic embryogenesis. The DNA segment located between −1048 and −792 bp (containing BBM and WUS REs) is needed for gene expression before embryogenesis onset but not during embryo development. The promoter segment between −792 and −618 bp (including GATA, ARR1AT, and ANT REs) regulates gene expression in developing embryos.

Abstract The cattle tick Rhipicephalus (Boophilus) microplus is an ectoparasite capable of transmitting a large number of pathogens, causing considerable losses in the cattle industry, with substantial damage to livestock. Over the years, important stages of its life cycle, such as the embryo, have been largely ignored by researchers. Tick embryogenesis has been typically described as an energy-consuming process, sustaining cell proliferation, differentiation, and growth. During the embryonic stage of arthropods, there is mobilization of metabolites of maternal origin for the development of organs and tissues of the embryo. Glycogen resynthesis in late embryogenesis is considered as an effective indicator of embryonic integrity. In the cattle tick R.(B. (B.) microplus, glycogen resynthesis is sustained by protein degradation through the gluconeogenesis pathway at the end of the embryonic period. Despite recent advancements in research on tick energy metabolism at the molecular level, the dynamics of nutrient utilization during R. (B.) microplus embryogenesis is still poorly understood. The present review aims to describe the regulatory mechanisms of carbohydrate metabolism during maternal-zygotic transition and identify possible new targets for the development of novel drugs and other control measures against R. (B.) microplus infestations.


Resumo O carrapato bovino Rhipicephalus (B.) microplus é um ectoparasita capaz de transmitir diversos patógenos, sendo responsável por grandes perdas na pecuária pelos danos causados ao gado. Atualmente, muitos estudos têm negligenciado fases importantes do ciclo de vida deste parasita, como a fase embrionária. A embriogênese é classicamente descrita como um processo que demanda um consumo de energia, possibilitando a proliferação celular, diferenciação e crescimento. Além disso, em artrópodes, o estágio da embriogênese é caracterizado pela mobilização de metabolitos de origem materna para o desenvolvimento de novos tecidos e órgãos. A ressíntese de glicogênio no final da embriogênese tem sido descrita em diversas espécies de artrópodes, sendo considerada um indicador de integridade do embrião. No caso do R. (B.) microplus a ressíntese de glicogênio é sustentada pela degradação de proteínas durante a gliconeogênese, no terço final da embriogênese. Apesar dos recentes avanços, no estudo molecular e do metabolismo energético, os mecanismos envolvidos na dinâmica da utilização de diferentes substratos energéticos durante a embriogênese do carrapato R. (B.) microplus ainda é pouco entendido. Diante deste panorama, estudos que descrevam a regulação destes mecanismos e da associação do metabolismo de carboidratos com a transição materno zigótica, pode auxiliar na busca de novos alvos para o desenvolvimento de novos acaricidas e outras intervenções para o controle infestações de R. (B.) microplus.

La importancia económica, ecológica y medicinal de las pasifloras ha fomentado el desarrollo de investigaciones multidisciplinarias en estas plantas. En cultivo in vitro, la embriogénesis no zigótica representa una alternativa para regeneración de plantas; no obstante, esta técnica ha presentado dificultades en la reproducibilidad de los protocolos, así como formación de ciertos embriones y plántulas anormales en algunas especies. En este estudio se evaluó la capacidad morfogénica de embriones zigóticos maduros (EZM) de Passiflora maliformis L para desarrollar embriones no zigóticos (ENZ), y posterior regeneración de plántulas. Se ensayó una etapa de inducción en la que se probaron 12 tratamientos en medio MS+VitB5 suplementado con 2,4-D, KIN y BA adicionados solos o combinados y, otra, de expresión en MS, MS con Carbón activado (CA) o MS con reguladores de crecimiento (RC) en concentraciones reducidas a 1/10 parte de las adicionadas en la etapa inducción. Altos porcentajes, 70%, de regenerantes fueron obtenidos en el medio suplementado con 1 mgL-1 BA y transferidos a medio de expresión MS, así como en el medio con 1 mgL-1 BA + 3 mgL-1 2,4-D, y transferidos a medio de expresión con CA, 60%. El 70% de las plántulas regeneradas se desarrollaron exitosamente en invernadero. Este trabajo es el primero que aborda la expresión del potencial embriogénico de EZM de P. maliformis induciendo exitosamente respuestas embriogénicas no zigóticas a través de rutas directas e indirectas; los resultados obtenidos son aplicables al conocimiento de la embriogénesis no zigótica en otras pasifloras, con fines de mejoramiento y uso comercial.


The economic, ecological and medical significance of passion fruits has encouraged the development of multidisciplinary research in this group. In vitro culture, non-zygotic embryogenesis represents an alternative for plant regeneration; however, this technique has presented difficulties in the reproducibility of protocols as well as the development of certain embryos and abnormal plantlets in some species. We assessed in Passiflora maliformis L the morphogenic capacity in the mature zygotic embryos (MZE) to develop nonzygotic embryos (NZE), and subsequent plantlets regeneration. We tested an induction stage with 12 treatments in MS + VitB5 medium supplemented with 2,4-D, KIN and BA added alone or combined and, another with a MS, MS with Activated Charcoal (AC) or MS with growth regulators (GR) in 1/10 concentrations reduced to 1/10 of those added in the induction stage. We obtained high percentages, 70%, of regenerants in the medium supplemented with 1 mgL-1 BA and transferred to the MS expression medium, as well as in the medium with 1 mgL-1 BA + 3 mgL-1 2,4-D, and transferred to the expression medium with AC, 60%. 70% of the regenerated seedlings were successfully grown in the greenhouse. This is the first research that addresses the expression of the MZE embryogenic potential in P. maliformis, inducing a successfully embryogenic non-zygotic responses through direct and indirect routes; the obtained results are relevant to the knowledge of non-zygotic embryogenesis in other passion fruits, for breeding and commercial uses.

The genus Citrus contains numerous fresh and processed fruit cultivars that are economically important worldwide, many genotypes are amenable to somatic embryogenesis, which became a key regeneration pathway in many experimental approaches of cultivar improvement. in this objective We aime at studying the effects of various culture media on the induction and the development of citrus somatic embryos.Callus cultures were initiated from the infertlized ovules of six varieties of mandarin (Anana, Lee, Murcott, Ortanique, Temple, and Wilking) within 3 media: MT (Murashig and Tuker, 1969) without hormones, MT + 1 mg/l BAP, MT + 1 mg/l Kinetin, the experiments show a highly significant effect (P < 0.001) of the culture media and genotype. No reactivity was observed on the MT environment in the absence of growth regulator, while the culture media MT in addition to 1 mg/l BAP gave the best results of induction of embryogenic callus induction. The induction of somatic embryogenesis was obtained on MT media without hormones. For the plantlets regeneration the favorable media was MT without hormones or added to ANA and active coal.
